ACH/AHS Application of Mass Spectrometry

Lecturer: Karel Lemr
Lecture: 1 hour/week + seminar 1 hour/week
Credits: 4
Winter semester
Form of course completion: colloquium

Introduction, an application of mass spectrometry in organic analysis, history and development. Structural analysis of small organic molecules (structure verification, identification of impurities in pharmaceutical substances, identification of metabolites), determination of molecular mass and elemental composition, fragmentation after electron ionization, collision induced dissociation, fragmentation pathways and a study of fragmentation. Application of ESI-MS, an optimization of ion source, an analysis of small volumes of a sample using nano-ESI. MALDI-TOF in an analysis of polymers, practical problems of ionization. Hyphenation of mass spectrometry and separation techniques. GC/MS in a toxicological analysis (databases of spectra). HPLC/MS – an optimization of analytical methods, limits of used interfaces and their comparison (e.g. ESI vs. APCI). HPLC/MS in an environmental and pharmaceutical analysis. CE/MS – implementation of a method, selected applications. Mass spectrometry of peptides and their fragmentation (sequence of amino acid). Application of FTICR. Advances in mass spectrometry. Discussion of mentioned topics in a seminar and possible solution of corresponding analytical tasks (e.g. interpretation of spectra of small organic molecules, data evaluation).
