Historical aspects. Basic properties of enzymes as catalysts. Cofactors. Nomenclature and classification of enzymes. Localisation and purification of enzymes. Structure of enzymes. Methods of study of active site. Covalent modification of enzymes. Mechanisms of enzyme actions. Enzyme kinetics – the Michaelis-Menten equation, Michaelis constant, reaction order, enzyme units, turnover number. Effect of pH on enzyme activity. Effect of temperature on enzyme activity. Enzyme reaction with one, two or three substrates. Enzyme specificity and regulation – allosteric regulation, covalent modification. Enzyme inhibition. Methods of determination of enzyme activity. Immobilization of enzymes. Control of metabolic pathways. Organized enzyme systems. Enzymes in the cell. Clinical aspects of enzymology. Enzyme technology.
